IAVI Starts First-in-Human Phase I Clinical Trial of Single-Dose Sudan Virus Vaccine Candidate

NEW YORK, NY / ACCESSWIRE / June 27, 2023 / IAVI announced today that the first participants have been vaccinated with a Sudan virus (SUDV) vaccine candidate in a first-in-human Phase I clinical trial in the U.S. The IAVI-sponsored trial is funded by the Biomedical Advanced Research and Development Authority (BARDA), part of the Administration for Strategic Preparedness and Response (ASPR) within the U.S. Department of Health and Human Services (HHS).

Known as IAVI C108, this first-in-human study is designed to evaluate the safety and immunogenicity of an investigational SUDV vaccine candidate donated to IAVI by Merck (known as MSD outside the U.S. and Canada) to supplement IAVI’s ongoing SUDV vaccine development program. This investigational SUDV vaccine candidate was produced for IAVI from existing investigational bulk drug substance previously manufactured by Merck. IAVI is the vaccine candidate’s regulatory sponsor. IAVI is responsible for all aspects of the candidate’s future development, including demonstrating equivalence between this SUDV vaccine candidate and IAVI’s other SUDV vaccine candidate, which utilizes the same viral vector but is manufactured using a new production platform.

The SUDV vaccine candidate being evaluated in IAVI C108 uses the same recombinant vesicular stomatitis virus (rVSV) viral vector platform as ERVEBO®, Merck’s single-dose Zaire ebolavirus (ZEBOV) vaccine which is licensed in the U.S., U.K., European Union, Canada, Switzerland, and 10 African countries. rVSV is also the platform technology utilized in IAVI’s portfolio of emerging infectious disease (EID) candidates.

IAVI C108 will take place at two U.S.-based clinical trial sites, where the study vaccine will be administered intramuscularly at three dosage levels. This is a placebo-controlled, single-blind study, meaning that only the researchers conducting the study will know whether a participant has received a vaccine dose or a placebo dose until after the trial is over. Approximately 36 healthy adults will be enrolled and will be followed for six months after vaccination to monitor their safety and immune responses to the vaccine candidate.

Like ZEBOV, SUDV is responsible for recurring viral hemorrhagic fever outbreaks, which have had lasting impacts on health security and geopolitical stability across sub-Saharan Africa. The estimated case fatality ratios of SUDV disease have varied from 41% to 100% in past outbreaks[1]. Despite high morbidity and mortality, there are no vaccines or therapeutics licensed for the prevention and treatment of SUDV. Existing ZEBOV vaccines and treatments are not effective against SUDV. Data being generated from ongoing studies suggest that rVSV could be a safe and effective platform for responding to SUDV and other related pathogens[2].

IAVI’s rVSV-based EID portfolio includes a SUDV vaccine candidate supported by BARDA; a Lassa fever virus vaccine candidate currently in a Phase I trial and supported by the Coalition for Epidemic Preparedness Innovations and the European & Developing Countries Clinical Trials Partnership; a Marburg virus vaccine candidate supported by BARDA and the Defense Threat Reduction Agency (DTRA) of the U.S. Department of Defense (DOD); and an intranasal SARS-CoV-2 vaccine candidate supported by DOD DTRA and the Japan Ministry of Finance.

The rVSV platform has been used extensively in adults and children[3]. The underlying vesicular stomatitis virus (VSV) is a common animal virus that does not cause serious illness in humans and has been investigated extensively as a vaccine vector. In the vaccine platform, it is engineered to encode a surface protein from a target pathogen – in this case, SUDV – to prompt the body to mount an immune response.

Much of the research and development on IAVI’s rVSV platform is performed at the IAVI Vaccine Design and Development Lab (DDL) in Brooklyn, New York. The DDL is located at the bioscience center in the historic Brooklyn Army Terminal. Since its founding in 2008, the IAVI DDL has become one of the world’s leading viral vector vaccine research and development labs, known for innovation and generation of novel vaccine design concepts.

Scientists with IAVI’s Human Immunology Laboratory (HIL) in London, U.K., are involved in processing participant samples and developing the analytical assays needed to evaluate IAVI C108 participants’ immune responses. Established in 2001, the HIL is fully integrated into the Infectious Disease department within the Faculty of Medicine at Imperial College London and serves as the clinical immunology reference laboratory for IAVI and the network of clinical research centers with which IAVI collaborates worldwide.

African Union calls for joint effort to peace in DRC

The Head of State of Comoros and acting president of the African Union (AU) Azali Assoumani Tuesday in Luanda called for joint efforts to address the situation in the DRC in order to avoid negative repercussions on other countries in the region.

Azali Assoumani made the call while delivering his speech at the Summit of Heads of States and Governments of the political and economic regions of Africa.

Addressing the event, aimed to coordinate efforts to ensure peace In the African continent, in particular the eastern region of the DRC, the Statesman asked for the need to step up cooperation in favour of the DRC.

He also spoke of the need to adopt effective political solutions necessary, to avoid duplication of efforts and competition between regional mechanisms.

In his speech, he suggested the creation of a coordination group composed of representatives of the AU, regional communities and the UN, within the framework of the pacification of the east of the DRC,

In turn, the president of the African Union Commission, Moussa Faki, defended engagement in the search for African solutions to the continent’s problems.

He endorsed the need to coordinate efforts for the efficiency and wise management of that country’s resources within the framework of engagements for the establishment of peace, through close and harmonious cooperation among the regional blocs.

Moussa Faki condemned the killing of civilians on the Uganda-DRC border.

He called for vigorous action to combat violations of international human and humanitarian rights.

The inter-regional meeting is among the deliberations of the 11th extraordinary summit of heads of state of the ICGLR held on March 3 in Luanda under the initiative of the Angolan president, Joao Lourenço.

The Summit is attended by Presidents Ali Bongo of Gabon, Évariste Ndayishimiye of Burundi, Félix Tshisekedi of DRC, Faustin-Archange Touadéra of Central African Republic and Emmerson Mnangagwa of Zimbabwe.

President of the Union of the Comoros and incumbent head of the African Union Azali Assoumani, Head of State of Zimbabwe and incumbent head of the African Union Peace and Security Council Emmerson Mnangagwa, President of Gabon, and incumbent head of the ECCAS Ali Bongo are all taking part in the quadripartite meeting.

Government to resolve complaint of Angola Post Office

The minister for Telecommunications, Information Technology and Media, Mário Oliveira, said on Tuesday in the Province of Luanda that the government will continue making efforts to meet the demands from the complaint book of the workers of the National Company of Postal Services and Telegraphs of Angola.

Speaking to the press, the minister said that negotiations are underway with the Board of Directors of the company’s trade union committee, stating that dialogue is the best way to put an end to the stand-off.

The government official called for the workers of the Angolan Post Office to be committed to work so that the company can make its effective contribution to the national economy.

“The Ministry is committed to the leverage of the company”, he said.

Workers from the National Company of Postal Services and Telegraphs of Angola have been on strike since 5 June, demanding an increase of wages and the application of occupational qualifications.

With roughly 400 workers, the state-owned company is supervised by the Ministry of Telecommunications, Information Technologies and Media.

Switzerland accredits new Angolan ambassador

The Angolan ambassador, Maria Filomena Delgado, Tuesday in Bern City presented to the President of the Swiss Confederation, Alain Berset, her Credentials for accreditation as Angola’s Extraordinary and Plenipotentiary Ambassador to that European country.

According to a note from the Embassy of Angola in Switzerland, during the audience granted to the diplomat by the Swiss President, ambassador Maria Filomena Delgado shared key information on the political, social and economic transformations underway in Angola.

At the time, the diplomat expressed her willingness to strengthen relations of friendship and cooperation in different areas between the two countries.

Search to begin in France for 47 German POWs executed in WWII

The search for 47 German soldiers killed in a mass execution in World War II, was to commence on Tuesday in a forested area in southern France. The investigation was prompted by a former French resistance fighter, who broke his silence about the shooting of German prisoners of war near the village of Meymac in June 1944, when the country was occupied by Nazi Germany. The executions came after German Waffen-SS troops, carried out a massacre in Tulle, to the south-east of Meymac, and obliterated the village of Oradour-sur-Glane, a war crime that became a symbol of Nazi barbarism in France. For decades it was generally known, that the German POWs and a French woman, accused of collaboration were shot dead. But all those involved had remained silent about the circumstances until the last surviving witness, a 98-year-old, spoke to local media in May. According to his account, those who were killed are buried in two mass graves. One with 11 bodies is said to have been located in a discrete operation in 1967, but the exhumation effort was halted. The other dead are said to be buried about 100 metres away. Broadcaster France 3 said investigators, will first use ground radar to find the graves, an effort that could take several days. If the Germans are found at the presumed site, the German War Graves Commission will arrange for their exhumation and burial in a German military cemetery, the French Defence Ministry said.
